#1 - Hepatic encephalopathy secondary to poisoning by Tephrosia cinerea in sheep

Abstract in English:

In the semiarid region of northeastern Brazil, Tephrosia cinerea causes periacinar hepatic fibrosis in sheep with severe ascites and, occasionally, nervous signs. Sixteen sheep from six outbreaks of T. cinerea poisoning were studied. All sheep had histologic lesion of periacinar fibrosis and six showed, in the brain, vacuolization (spongy degeneration) of the white matter and junction between grey and white matter and presence of Alzheimer type II astrocytes in the grey matter. The disease was produced experimentally in two sheep, that presented porto-sistemic shunts and similar histologic lesions as those observed in the spontaneous cases. Immunohistochemistry revealed weak labelling with anti-GFAP antibodies suggesting a degenerative alteration of astrocytes with accumulation of dense bodies and reduction of the GFAP. There was strong labelling with anti-S100 antibodies suggesting cellular reactivity with proliferation of mitochondria and endoplasmatic reticulum. Such alterations are characteristic of the effects caused by ammonia on the astrocytes. It is concluded that in poisoning by T. cinerea nervous signs due to hepatic encephalopathy occur in some sheep.

Bento Gonçalves 9090, Bairro Agronomia, Porto Alegre, RS 91540-000, Brazil. E-mail: angelicawouters@yahoo.com.br Senecio spp. poisoning is an important cause of illness and death of cattle in Rio Grande do Sul, Brazil, and is often manifested by neurologic clinical signs and histological brain changes. Histological evaluation was performed on liver and brain samples of ten cattle naturally poisoned by Senecio sp. Samples of cerebrum, brainstem and cerebellum were stained with periodic acid-Schiff (PAS), and immunohistochemistry was carried out, employing anti-fibrillary acidic protein (GFAP), anti-S100 protein and anti-vimentin antibodies. The histological finding in the brain included mild to severe vacuolation in the white matter and the junction of gray and white matter, characterized as spongy degeneration. Histochemical and immunohistochemical staining revealed no significant findings compared with the brains of eleven adult cattle without liver and/or brain changes used as controls.

E-mail: a2ag@cav.udesc.br Two outbreaks of closantel overdosage in sheep and goat flocks are described. In the first outbreak 12 sheep were affected, 5 of them showed blindness, three sheep died (Sheep 1-3) and two were euthanized 6 months after the onset of clinical manifestation (Sheep 4 and 5). In the second outbreak 26 goats were affected, from which six survived despite blindness and one was euthanized. Clinically the animals showed depression, ataxia, motor incoordination, decreased or absent pupil reflexes. In some animals this clinical picture developed to bilateral blindness, with no reaction to threat and bilateral irresponsive midriasis. In the ophthalmic examination retinal vessel atrophy and hyperreflexia were observed. The histological examination showed myelin edema leading to status spongiosus in the central nervous system and compressive neuropathy of the optic nerve, associated with retinal degeneration and/or atrophy. This report aims to describe the epidemiologic, clinic and pathologic aspects of closantel overdosage in sheep and goats.

E-mail: alschild@terra.com.br To determine the reversibility of neurological and cardiac lesions in Ateleia glazioviana and Tetrapterys multiglandulosa poisoning, 3 groups of four sheep each were fed orally with the plants. In Group 1, 2 and 3, when sheep with nervous signs showed considerable increase of symptoms and risk of death, the plant administration was suspended. Group 4 with two sheep was used as control. Sheep from Group 1, fed 10g/kg during 6 days of fresh A. glazioviana collected in autumn were euthanized 8, 11, 16, and 21 days after the start of the experiment; they had shown regression of nervous signs, but had progressive cardiac lesions. Sheep from Group 2, fed during 8 days the same dose of fresh A. glazioviana collected in spring, were euthanized on days 9, 23, 38 and 68; they had shown regression of nervous signs and had no cardiac lesions. This suggests that the plant is less toxic in spring. Sheep from Group 3, fed 10g/kg during 11 days dry and ground T. multiglandulosa mixed with concentrated food, were euthanized on days 33, 33, 92 and 92; they had shown regression of nervous signs, and cardiac lesions were less severe on day 33 than on day 92. These results indicate that nervous lesions are reversible after the end of feeding, but cardiac lesions are progressive after discontinuation of the plant administration; these can be afterwards reversible if the animals do not anymore show clinical signs or die as a consequence of the poisoning. Results of those and previous experiments show that lesions of the nervous system are induced by lower doses than cardiac lesions and occur within a shorter period, what suggests that in spontaneous cases cardiac signs are always preceded by nervous signs, and that nervous signs can occur in the absence of cardiac lesions.

Abstract in English:

ABSTRACT.- Guedes K.M.R., Schild A.L. Riet-Correa F., Barros S.S. & Simões S.V.D. 2006. [Spongious degeneration in the central nervous system in Sindhi calves]. E-mail franklin.riet@pesquisador.cnpq.br Spongy degeneration (status spongiosus) includes many histological alterations characterized by vacuoles in the neuropile in different regions of the central nervous system (CNS). The vacuolization can be caused by intramyelinic edema, as in maple syrup urine disease and some toxic diseases, or by astrocytic edema, as in citrullinemia. This paper reports a spongy degeneration in two Sindhi calves, a female and a male. Both calves were offspring from the same bull, and the herd had high degree of inbreeding. The female calf was born normal, but progressive nervous signs were observed since 2 months old. The male calf had progressive nervous signs since it was born. Both calves were euthanized with severe nervous signs when 4 months old. At necropsy the liver was pale in the male calf. Histological alterations were mild to accentuated vacuolization, mainly in the deep cerebral cortex, cerebellar white matter, grey matter of the spinal cord, internal capsule, and brain stem. Fatty degeneration was observed in the liver of both animals. On electron microscopy it was found that the status spongiosus was caused by astrocytic edema. It is suggested that the disease is caused by a metabolic hereditary error.

Abstract in English:

Riet-Correa G., Terra F.F., Schild A.L., Riet-Correa F. & Barros S.S. 2005. [Experimental poisoning by Tetrapterys multiglandulosa (Malpighiaceae) in sheep.] E-mail: griet@ufpa.br Cardiac fibrosis was observed in a calf showing dullness, weakening and respiratory insufficiency in a farm in the state of São Paulo, where cardiac insufficiency, abortion and nervous signs in cattle were associated with the ingestion of Tetrapterys multiglandulosa. The objectives of this paper were to determine the susceptibility of sheep to the intoxication by T. multiglandulosa, to describe the clinical and pathological characteristics of the intoxication, and to evaluate the possibility of using sheep as an experimental species for toxicological studies with this plant. In a previous experiment to determine the toxicity of T. multiglandulosa to be used in sheep, the green plant was given to a steer at the dose of 22g per kg body weight (g/kg/bw), daily, during 9 days. After 9 days the steer showed nervous signs, and on the 12thday was euthanatized. No gross lesions were observed at necropsy. Status spongiosus was observed on the deeper layers of the cerebral cortex and subcortical white matter. Six male sheep were divided in three groups of two sheep each. Group 1 (Sheep 1 and 2) received daily doses of 6 g/kg/bw of the dry plant, during 30 days; Group 2 (Sheep 3 and 4) received daily doses of 3 g/kg/bw, during 60 days; and Group 3 was the control group. Sheep 1 was euthanatized 30 days after the start of the ingestion. Only cardiac arrhythmia was observed clinically, and no lesions were observed at necropsy. Sheep 2, 3 and 4 had also cardiac arrhythmia from day 9, 12 and 18, respectively. From day 52 they started to show depression, reluctance to move and incoordination. Clinical signs got gradually worst and the sheep were euthanatized on days 60, 70 and 80, when clinical signs were marked, and the animals will die at any moment. Hydrothorax, hydropericardium, ascites, nutmeg appearance of the liver and hard whitish myocardium, mainly in the interventricular septum and left ventricle, were observed at necropsy. Histologically, the heart of Sheep 2, 3 and 4 had areas of fibrosis associated with mononuclear cell infiltration. No lesions were found in the heart of Sheep 1. The 4 treated sheep had status spongiosus in different areas of the cerebrum and brain stem, mainly of the deep layers of the cerebral cortex and subcortical white matter. Status spongiosus were also observed in the cerebellar white matter and cervical medulla. In the later the vacuolization was moderate in the white matter and mild in the grey matter. Status spongiosus were mild in Sheep 1 and moderate to severe in Sheep 2, 3 and 4. On electron microscopy it was observed that the status spongiosus is due to an intramyelinic edema. No gross or histologic lesions were observed on the two control sheep, which were euthanized on day 80 after the start of the experiment.

#7 - Experimental poisoning by Ateleia glazioviana (Leg.Papilibnoideae) in sheep, 21(3):98-108

Abstract in English:

ABSTRACT.- Stigger A.L., Barros C.S.L., Langohr I.M. & Barros S.S. 2001. [Experimental poisoning by Ateleia glazioviana (Leg.Papilibnoideae) in sheep] Intoxicação experimental por Ateleia glazioviarya (Leg.Papilionoideae) em ovinos. Pesquisa Veterinária Brasileira 21(3):98-108. Depto Patologia, Universidade Federal de Santa Maria, 97105-900 Santa Maria, RS, Brazil. E-mail: cslb@sm.conex.com.br The leaves of Ateleia glazioviana were fed daily to seven sheep. Another sheep was used as côil(rol,. Total amounts (g/kg/body weight) o(;the plant consumed by each animal were 75 during 60 days (Sheep 6), 125 during-25 days (Sheep 4), 13,0 during 13 days (Sheep 7), 150 during 60 days (Sheep 5), 180 during 18 days (Sheep 3), 330 during 33 days (Sheep 2), and 375 during 25 days (Sheep 1 ). Six sheep were fed the green leaves while Sheep 7 was fed dried A. glazioviana leaves. Five sheep (Sheep 1-4 and 7) developed clinical signs 8 to 16, days after the first administration of the plant. The clinical course varied from 6 to 50 days and clinical signs included apathy, loss of appetite, and increased heart and respiratory rates; affected sheep were ·reluctant to move and showed a low carriage of the head, instability of the hind limbs, and staggering gait. Some sheep supported their weight against the wall of the stable. Sheep 1, 3, and 4 died spontaneously; paddling movements preceded death. Sheep 2 and 7 were euthanatized when presenting marked clinical signs of the toxicosis. Sheep 5 and 6 did not get clinically ill and were euthanatized along with the control sheep 103 days after the first administration of the plant. All sheep were necropsied and several of their organs, including heart and brain, were evaluated histologically. Additionally, fragments of the myocardium of Sheep 3 and 4 were evaluated by transmission electron microscopy. To provide more controls for the myocardial changes, the hearts were collected from six animals at an abattoir known to slaughter sheep from regions free of A. glazioviana. These hearts were processed for histology and examined in the same way as the other eight. All seven treated (plant fed) sheep had gross changes at necropsy. There were firm, irregular white or yellow areas in the myocardium of all treated sheep. These pale areas were more evident on the cut surface of the heart. Hydrothorax and/or hydropericardium were observed in five animals (Sheep 1-4 and 7). Nutmeg livers were seen in Sheep 1 and 7. This latter sheep also had ascites. The main histopathological findings in treated sheep were degenerative/necrotic changes in the myocardium. The early changes consisted of acute swollen myocardial fibers associated with bizarre nuclei. Apparently this change progressed until the sarcoplasm disappeared and an empty collapsed sarcolemmal tube remained surrounded by interstitial connective tissue and fibrosis. Overt necrosis (hyaline and floccular) associated with mono nuclear infiltration and phagocytosis of fiber fragments were also observed. Varying degrees of spongy degeneration were seen in the white matter of the brain of Sheep 1, 3, 4 and 7. Mitochondrial swelling and loss of density of the mitochondrial matrix were the earliest ultrastructural changes observed. In advanced lesions cardiac cells had vacuoles in the sarcoplasm, rupture of the sarcolemma and necrosis with intervening macrophages and fibroblasts associated with increased collagen.

RESUMO.- Gava A., Barros C.S.L., Pilati C., Barros S.S. & Mori A.M. 2001. [Poisoning by Ateleia glazioviana (Leg. Papilionoideae) in cattle] Intoxicação por Ateleia glazioviana (Leg.
